New lifesaving antibiotic to fight sepsis and pneumonia enters final human testing phase

Zosurabalpin targets drug-resistant bacteria behind sepsis and pneumonia

A potential game-changer in the fight against deadly hospital infections is entering the final stage of human trials. Swiss pharmaceutical giant Roche announced it is moving forward with Phase 3 testing of its experimental antibiotic, zosurabalpin, a drug designed to combat highly drug-resistant infections that threaten millions each year.

Jointly developed with Harvard University, Zosurabalpin will be tested specifically against Acinetobacter baumannii — a dangerous pathogen that the U.S. Centers for Disease Control and Prevention (CDC) has labeled an “urgent threat” due to its resistance to current treatments. Notably, it’s been more than half a century since the last effective antibiotic was developed to treat infections caused by this microbe.

A silent killer: the global burden of sepsis and pneumonia

Hospital-acquired infections such as pneumonia and sepsis continue to exact a staggering human toll. Sepsis alone is responsible for 11 million deaths annually — roughly one person every 2.8 seconds — while community-acquired pneumonia (CAP) kills up to four million people each year, disproportionately affecting older adults with pre-existing health conditions.

Zosurabalpin’s success could mark a critical turning point in modern medicine’s battle against antimicrobial resistance (AMR), a growing threat that undermines decades of medical progress.

Global clinical trial begins with hope for end-of-decade approval

The Phase 3 clinical trial will enroll approximately 400 patients from diverse global locations, Roche said. The goal is to evaluate the efficacy and safety of the drug in treating severe infections where conventional antibiotics often fail.

“Our goal is to contribute new innovations to overcome antimicrobial resistance, one of the biggest infectious disease challenges to public health,” said Michael Lobritz, Roche’s global head of infectious diseases, in a statement to The Independent.

Larry Tsai, senior vice president and global head of immunology at Roche’s infectious disease unit, emphasized the scale of the crisis. “These drug-resistant bacteria are present in every country of the world,” Tsai noted. “The innovative biology involved in this research could potentially reveal new insights into the structure of bacterial membranes, possibly leading to the discovery of new antibiotics in the future.”

A potential milestone in the post-antibiotic era

Zosurabalpin’s development reflects renewed urgency in pharmaceutical innovation following years of stagnation in antibiotic research. Public health experts have long warned that without effective new antibiotics, the world could face a “post-antibiotic era” where routine surgeries and minor infections once again become fatal.

If successful, zosurabalpin could be approved for public use by the end of the decade, potentially saving millions of lives and ushering in a new era of antimicrobial treatment.
